Effects of erythropoietin on the neurogenesis of vascular dementia rats

文摘 目的观察促红细胞生成素(EPO)对血管性痴呆(VD)大鼠神经发生和认知功能的影响.方法 SD大鼠45只,按随机数字表达法分为对照组,VD组和治疗组;改良2-VO+硝普钠法建立VD大鼠模型;治疗组腹腔注射EPO;电脑控制的穿梭箱系统检测大鼠认知功能;免疫荧光法检测神经干细胞增殖情况;western blot测定脑内细胞因子表达情况.结果造模后30d行为学测试,VD组大鼠主动回避反应(AAR)比率显著低于对照组(P<0.01);治疗组大鼠AAR比率显著高于VD组;免疫荧光结果 :BrdU标记增殖细胞治疗组明显高于对照组和VD组.Western blot检测显示治疗组脑内脑源性神经营养因子(BDNF),血管内皮生长因子(VEGF)表达明显高于其余两组.结论腹腔注射EPO可促进脑内细胞因子BDNF,VEGF分泌及神经发生,并显著改善大鼠的认知功能
其他语种文摘 Objective To explore the effects of erythropoietin (EPO) on the neurogenesis and cognitive function of vascular dementia rat. Methods Sprague-Dawlev (SD) rats were randomly and equally divided into the sham control group, the VD group, and the treatment group. The VD rat model was established by the way of cerebral ischemia/reperfusion repeatedly and received intraperitoneal infusion sodium nitroprusside, the treatment group was treated with EPO, and the others were treated with saline. The learning-memory ability was measured by using computerized shuttle-training case and the neurogenesis was detected by immunofluorescence staining. And the expression of cytokine was observed by Western blot. Results The active avoidance reaction (AAR) ratio in the VD group was significantly lower than that in the sham control group (P<0.01), but that in the treatment group was higher than that in the VD group (P<0.01). Immunofluorescence staining showed that the number of BrdU labeled cell in the treatment group was significantly higher than that in the sham control group and the VD group. Western blot showed that the expression of BDNF and VEGF in the treatment group were significantly higher than that in the other group. Conclusion Treatment of erythropoietin could enhance neurogenesis, promote cytokine BDNF and VEGF secreting, and improve the ability of learning and memory in VD rats
