建立了高效液相色谱法测定不同油脂中脂肪醛的方法。各种油样品前处理后与2,4-二硝基苯肼(DNPH)发生衍生反应,在色谱条件为ODS-A色谱柱(5 μm,250 ×4.6 mm ID),流动相是乙腈和水(体积比为75∶25),以HAc调节pH为3,检测波长为356nm,流速为1.0 mL/min的条件下进行检测。测得6种脂肪醛的线性范围为1.0×10~(-7)~1.0×10~(-4) mol/L,检出限为3.2×10~(-8)~1.48×10~(-7)mol/L。实际样品加标回收实验,回收率为87.8%~113.3%,相对标准偏差为3.2%~7.8%。 |

A simple and effective method has been developed for sensitive determination of six aliphatic monoaldehydes, including formaldehyde, acetaldehyde, propanal, butanal, pentanal and hexanal in various oil samples. The oil samples were enriched by passing through a silica gel column and gradient elution. After being derivatized with 2,4-dinitrophenylhydrazine (DNPH), these six aliphatic aldehydes were separated and analyzed with in 35 min on a reversed phase ODS column with methanol/0.1% acetic acid water solution as the mobile phase at a flow rate of 1.0 mL/min. Experimental conditions of derivatization and HPLC detection were optimized. The proposed method was validated according to International Conference on Harmonization (ICH) requirements, with recovery results ranging from 87.8% to 113.3%. Calibration plots of aliphatic aldehydes were linear (r≥0.9991) in the concentration range from 1.0×10~(-7) to 1.0×10~(-4) mol/L. The LODs were between 3.2×10~(-8) and 1.48×10~(-7)mol/L. |
