采用高效液相色谱法测定桔梗中10种主要的皂苷类物质,桔梗样品经甲醇(60+40)溶液超声振荡提取后,过Waters Spe-Pak-C_(18)固相萃取柱富集浓缩,以10mL·min~(-1)的流速,用甲醇(60+40)溶液将皂苷类物质从固相萃取柱上洗脱下来.以ZORBAX Stable Bound C_(18)(4.6 mm×100 mm,1.8μm)为快速分离柱,水和乙腈为流动相梯度洗脱,用蒸发光散射检测器检测.峰面积与10种皂苷类物质的质量浓度呈线性关系,方法检出限(3S/N)在0.3~0.8 mg·L~(-1)之间.采用标准加入法做回收率试验,回收率在84%~106%之间,相对标准偏差(n=5)小于5%. |

HPLC was applied to the determinations of 10 main saponins in roots of balloon flower. The sample was extracted ultrasonically with a mixed solution of methanol and water (60+40), and enriched by passing through a Waters Spe-Pak-Qs SPE column, Saponins were eluted from the column with mixed solution of methanol and water (60+40) at a flowrate of 10 mL · min~(-1). The eluate was separated by gradient elution with acetonitrile solution of different concentration on ZORBAX Stable Bound C_(18 )(4.6 mm×100 mm, 1.8μm) rapid column, and their contents determined by evaporation light scattering detector. Linear relationships between the peak area and mass concentration of 10 saponins were obtained, with detection limits(3S/N)in range of 0.3—0.8 mg·L~(-1). The values of recovery found by standard addition method were in the range of 84% —106%, with valus of RSD's (n=5) less than 5%. |
