High Performance Liquid Chromatographic Determination of Docetaxel in Human Serum Albumin Nanoparticles by Addition of Cysteine

建立多西紫杉醇(docetaxel,DOC)人血清白蛋白纳米粒(humanserum albumin nanoparticles,HSA—NP)中药物含量测定的方法。利用半胱氨酸(cysteine,Cys)还原人血清白蛋白中交联的二硫键,释放HSA—NP中包裹的药物,再利用HPLC测定药物含量。结果表明,在本实验条件下Cys及制剂中辅料对DOC的含量测定无干扰。1μg/L Cys、37℃孵化30min,最后用乙腈沉淀蛋白的方法可以完全测定出制剂中DOC含量。 |

A method was established for the determination of docetaxel (DOC) in human serum albumin (HSA) nanoparticles by the addition of cysteine. The effect of cysteine (the disulphide bond breaker) on HSA disulphide bonds was monitored by the detection on the content of DOC from HSA-NP using HPLC. The measurements were performed on addition of 1 ~g/L free cysteine to the samples, the content assay of DOC were carried out after an incubation period of 30 min at 37 ~ under continuous shaking. The results showed that the addition of cysteine significantly increased the release of DOC from HSA-NP, and the determination results of DOC were 10% higher than that of acetonitrile. |
