目的:建立同时测定苦豆子中槐定碱、氧化槐果碱及氧化苦参碱含量的HPLC测定法。方法:色谱柱为PhenomenexGeminiC,8(4.6mm×250mm,5μm),以甲醇(A)-0.2%磷酸水(B)为流动相,检测波长205nm,流速1mL·min^-1,柱温30℃。结果:在线性范围内3个对照品的标准曲线呈良好的线性关系(r≥0.9997)。平均回收率分别为100.0%,97.0%,97.0%。结论:可通过HPLC同时测定苦豆子中槐定碱、氧化槐果碱和氧化苦参碱的.含量,并可用该方法进行苦豆子药材的质量控制。 |

Objective: To develop a HPLC method for simultaneous determination of the contents of sophoridine,oxymatrine and oxysophocarpine in seed of Sophora alopecuroides. Method: Chromatographic condition included a Phenomenex Gemini C18 (4- 6 mm ×250 mm, 5μm) column and thmobile phase consisting of a mixture of methanol-0. 2% phosphatic acid (7:93). The detection wavelength was at 205 nm. The flow rate was 1.0 mL · min^- 1 and column temperature was set at 30 ℃. Result: All calibration curves showed good linearity (r≥0. 999 7) within test ranges. The mean recoveries were 100. 0%, 97.0% and 97.0% respectively. Conclusion: This method is simple, rapid and accurate , and suitable for quality control of S. alopecuroides seed. |
