活动性系统性红斑狼疮患者CD8+T细胞CCR7异常高表达

目的探讨系统性红斑狼疮患者T细胞是否存在CCR7表达异常及功能改变.方法应用流式细胞仪、实时定量RT-PCR和Northern印记分别检测正常机体和SLE患者CD4+T细胞和CD8+ T细胞CCR3、CCR4、CCR5、CCR7和CCR9的表达情况;利用趋化实验检测其功能.结果活动性SLE患者CD8+T细胞异常高表达CCR7(94.8%),CD8+T细胞CCR7mRNA水平明显增高(拷贝数约5.3×103±1.1×103);CCL21可选择性诱导活动性SLE患者CD8+T细胞发生明显的趋化运动,最大C.I.值为3.8±0.42.结论 CCR7通常选择性、功能性地高表达于活动性SLE患者的CD8+T细胞上,并不异常表达于其CD4+T细胞,而该现象并不见于正常机体和非活动性SLE患者. |

Objective To determine whether differential expression and functions of the CCR7 occur in SLE patients. Methods Flow cytometry,real time quantitative RT-PCR and Northern blotting were used to detect expression and functions of CCR3 , CCR4, CCR5, CCR7 and CCR9 on CD4+ and CD8+ T cells from normal and SLE subjects;chemotaxis assay was used to detect the functions. Results CCR7 was abnormaly and highly expressed on CD8+ T cells from the active SLE patients(94. 8%) ;mRNA CCR7 obviously increased in the same cells (approximately 5. 3± 1. 1 X 103 copies) ;CCL21 selectively induced significant chemotactic migration of those cells ,the maximum C. I. was 3. 8 + 0. 42. Conclusion CCR7 is selectively,frequently and functionally expressed on CD8+ but not CD4+ T cells from active SLE patients,whereas this phenomenon would not have been seen in normal and inactive SLE subjects. |
