文摘 目的观察低频超声对人卵巢癌COC1体外生长的影响.方法以低频超声(频率0.24 MHz,剂量5.12W/cm~2*5s)辐照COC1细胞,采用MTT观察超声对COC1细胞增殖的抑制作用;流式细胞仪(FCM)、TUNEL法和光、电镜检测细胞凋亡.结果超声波能够显著抑制卵巢癌COC1细胞增殖,辐照后12h、24h、36h和48h,其MTT光吸收值与对照组相比显著降低;FCM检测超声辐照后6h的COC1细胞凋亡率达(25.57±4.68)%;同时TUNEL法观察到棕褐色着染的凋亡细胞;相差显微镜及电镜下可见凋亡小体形成等典型形态学改变.结论低频超声辐照对人卵巢癌COC1细胞体外生长具有明显的抑制作用,并可诱导其凋亡.

Methods The growth inhibitory effect of Ultrasound Exposure(0.24 MHz, 5.12 W/cm~2 * 5 s) on COCI cell was observed by MTT colormetric assay; Cell apoptosis was analyzed using flow cytometry (FCM); Terminal deoxynucleotidyl transferase-mediated dUTP-digoxin nick end labeling (TUNEL); Phase contrast light microscope and Electron microscope were applied to detect the morphological changes of COCI cell. Results Sonication could obviously inhibit the proliferation of COCI cell; The absorbance value(As7o)of MTT significantly decreased at 12h,24h,36h and 48h after sonication as compared to control group; The apoptosis rate was found to reach (25. 57 ± 4.68) % by FCM, and brown-colored positive apoptotic cells were observed with TUNEL method at 6h after sonication; Typical morphological changes of apoptosis such as apoptotic body were found under Phase contrast light microscope and Transmission electron microscope. Conclusion The results suggest Low-Frequency Ultrasound can suppress proliferation and induce apoptosis in vitro in human ovarian cell line COCI.
